RESUMO

Electrophorus spp. generate high-voltage electric discharges for defense and hunting, and low-voltage electric discharges (as other Gymnotiformes) for electrolocation and communication. Despite intense interest in the unusual electrogenic and electroreceptive capacities of electric eels, the other sensory systems of Electrophorus spp. are relatively poorly known. Here we describe the ontogenetic development and organization of the cephalic lateral-line canals in the lowland electric eel, Electrophorus varii. Preserved specimens of larvae, juveniles, and adults were examined to describe the spatial distribution of the canals and pores. Ontogenetic shifts of the cephalic lateral line formation were observed for each canal and support a hypothesis of non-synchronized development. The morphogenesis of cephalic canals in larvae and juveniles begins just before the onset of exogenous feeding. In adults, the cephalic sensory canals are formed separately from the skull and overlay cranial and mandibular bones and muscles. This study provides the first detailed description of the development and organization of the cephalic lateral-line system in Electrophorus varii.(AU)


Electrophorus spp. geram descargas elétricas de alta voltagem, usadas para defesa e caça, e (como os demais Gymnotiformes) descargas elétricas de baixa voltagem para eletrolocalização e comunicação. Apesar do grande interesse nas capacidades eletrogênicas e eletrorreceptivas incomuns dos poraquês, os outros sistemas sensoriais de Electrophorus spp. são relativamente pouco conhecidos. Aqui nós descrevemos o desenvolvimento ontogenético e a organização dos canais da linha lateral cefálica no poraquê da planície Electrophorus varii. Exemplares preservados de larvas, juvenis e adultos foram analisados para descrever a distribuição espacial dos canais e poros. Variações ontogenéticas na formação da linha lateral cefálica foram observadas para cada canal, apresentando um desenvolvimento não-sincronizado. Variações ontogenéticas da formação da linha lateral cefálica foram observadas para cada canal e suportam uma hipótese de desenvolvimento assincrônico. A morfogênese dos canais cefálicos em larvas e juvenis precede o início da alimentação exógena. Nos adultos, os canais sensoriais cefálicos se formam separadamente do crânio e sobrepõem os ossos e músculos cranianos e mandibulares. Este estudo fornece a primeira descrição detalhada do desenvolvimento e organização do sistema da linha lateral cefálica de Electrophorus varii.(AU)

INTRODUCTION: This study aimed to describe the profile of freshwater stingray injuries in the State of Amazonas, Brazilian Amazon, and to identify the associated risk factors for secondary infections. METHODS: This cross-sectional study used surveillance data from 2007 to 2014 to identify factors associated with secondary infections from stingray injuries. RESULTS: A total of 476 freshwater stingray injuries were recorded, with an incidence rate of 1.7 cases/100,000 person/year. The majority of injuries were reported from rural areas (73.8%) and 26.1% were related to work activities. A total of 74.5% of patients received medical assistance within the first 3 hours of injury. Secondary infections and necrosis were observed in 8.9% and 3.8%, respectively. Work-related injuries [odds ratio (OR) 4.1, confidence interval (CI); 1.87-9.13] and >24 hours from a sting until receiving medical care (OR; 15.5, CI; 6.77-35.40) were independently associated with the risk of secondary bacterial infection. CONCLUSIONS: In this study, work-related injuries and >24 hours from being stung until receiving medical care were independently and significantly associated with the risk of secondary infection. The frequency of infection following sting injuries was 9%. The major factor associated with the risk of secondary bacterial infection was a time period of >24 hours from being stung until receiving medical care.

Porcine circovirus (PCV) is a potentially harmful virus that has been shown to contaminate biological products. The virus is resistant to many inactivation and/or removal procedures performed during manufacturing. Anion exchange chromatography has been shown to be useful for PCV type 1 (PCV1) removal; however, reduction of PCV1 using methods such as heat inactivation, low pH, nanofiltration, UV-C, and gamma irradiation has not been successful. Therefore, in this study, we evaluate various conditions for process solutions during nanofiltration using PCV1. The results indicated that PCV could be effectively removed from glycine solution at 0.1-0.3 M, pH 4.0 without IgG, using a nanofilter with a pore size of 19 nm (19-nm filter); log reduction values (LRVs) of ≥4.5 and ≥ 5.0, respectively, were obtained. In contrast, PCV1 was significantly removed (LRV: 2.2) in glycine solution at 0.3 M, pH 6.0 with 1.0% IgG using the 19-nm filter, but some virus genomes were detected in the filtrates. In summary, the use of a 19-nm filter in glycine solution with/without IgG is an appropriate condition for PCV removal.

Emerging X-ray free-electron lasers with femtosecond pulse duration enable single-shot snapshot imaging almost free from sample damage by outrunning major radiation damage processes. In bioimaging, it is essential to keep the sample close to its natural state. Conventional high-resolution imaging, however, suffers from severe radiation damage that hinders live cell imaging. Here we present a method for capturing snapshots of live cells kept in a micro-liquid enclosure array by X-ray laser diffraction. We place living Microbacterium lacticum cells in an enclosure array and successively expose each enclosure to a single X-ray laser pulse from the SPring-8 Angstrom Compact Free-Electron Laser. The enclosure itself works as a guard slit and allows us to record a coherent diffraction pattern from a weakly-scattering submicrometre-sized cell with a clear fringe extending up to a 28-nm full-period resolution. The reconstructed image reveals living whole-cell structures without any staining, which helps advance understanding of intracellular phenomena.

In spite of the fact that catfish have rarely been reported as food items in the diets of freshwater stingrays, they are commonly observed in the stomach contents of species inhabiting the Solimões and Negro rivers in the Brazilian Amazon. The stomach contents of six rays from the Solimões River and 64 from the Negro River were analyzed, and catfishes (distributed among the families Callichthyidae, Cetopsidae, Doradidae and Loricariidae) were found in the stomach contents of four and 10 of these rays, respectively, comprising a frequency of occurrence of 20%. These data indicate a relevant participation of catfishes in the diets of potamotrygonid rays in the Amazon, and may reflect the regional diversity and abundance of Siluriformes in the region.


Apesar de ter sido reportado sobre a raridade de bagres na alimentação de raias de água doce, a presença deles tem sido comumente observada em conteúdos estomacais de espécies dos rios Solimões e Negro. Os conteúdos estomacais de seis indivíduos de raias do rio Solimões e 64 do rio Negro foram analisados e os bagres (distribuídos nas famílias Callichthyidae, Cetopsidae, Doradidae e Loricariidae) foram consumidos por quatro e 10 raias, respectivamente, com a frequência de ocorrência de 20%. A ocorrência de Siluriformes, mesmo baixa, indica uma participação relevante deste grupo como presas na dieta dos potamotrigonídeos.

The feeding mechanisms of elasmobranchs and their functioning have been receiving growing scientific attention, although less emphasis has yet to be directed towards batoid species. The present study investigated the use of orobranchial musculature during prey capture and processing behavior in Potamotrygon motoro. Ten orobranchial muscle groups were removed to determine their relative biomasses. The kinematics of the musculature was described based on videos of prey capture and manipulation under captive conditions recorded at 250 and 500 field/s. Analyses of the orobranchial musculature indicated that adductor muscles responsible for closing the mouth had high biomasses, which is consistent with their functions related to apprehend and manipulate the prey. The feeding behavior adopted by this ray species showed a sequence of manipulation to crush hard prey as crustaceans (dominant prey category for P. motoro) to facilitate final oral transport. The morphological and behavioral characteristics of P. motoro are essential to fully understanding the mechanisms used in prey capture and processing in environments in which these stingrays occur.


Os mecanismos alimentares e suas funções em elasmobrânquios têm recebido crescente atenção, apesar de pouca ênfase ser direcionada às espécies de batoideos. No presente estudo foi investigado o uso da musculatura orobranquial na captura e processamento de presas durante o comportamento alimentar em Potamotrygon motoro. Dez conjuntos de músculos orobranquiais foram removidos para a determinação da massa relativa. A cinemática dos músculos foi descrita a partir de observações da captura e manipulação de presas registradas em 250 e 500 fotogramas/s, sob condições de cativeiro. A análise da musculatura orobranquial indicou que os músculos adutores responsáveis pelo fechamento da boca apresentaram alta biomassa, o qual foi consistente com sua função para a apreensão e manipulação de presas. O comportamento alimentar utilizado por esta raia mostrou uma sequência de manipulação para a quebra de presas com estruturas rígidas como crustáceos (considerados como categoria dominante de presas para P. motoro), facilitando o transporte oral final. As características morfológicas e comportamentais de P. motoro são essenciais para a compreensão do uso do mecanismo para a captura e processamento de presas nos ambientes em que essas raias ocorrem.

The relationship between the distribution of the lateral line canals and their functionality has not been well examined in elasmobranchs, especially among Neotropical freshwater stingrays of the family Potamotrygonidae. The spatial distribution of the canals and their tubules and the quantification of the neuromasts were analyzed in preserved specimens of Potamotrygon motoro, P. orbignyi, Potamotrygon sp. "cururu", and Paratrygon aiereba from the middle Negro River, Amazonas, Brazil. The hyomandibular, infraorbital, posterior lateral line, mandibular, nasal and supraorbital canals were characterized and their pores and neuromasts quantified. The ventral canals are known to facilitate the accurate localization of prey items under the body, and our results indicate that the dorsal canals may be employed in identifying the presence of predators or potential prey positioned above the stingray's body. The presence of non-pored canals in the ventral region may be compensated by the high concentration of neuromasts found in the same area, which possibly allow the accurate detection of mechanical stimuli. The concentration of non-pored canals near the mouth indicates their importance in locating and capturing prey buried in the bottom substrate, possibly aided by the presence of vesicles of Savi.


A relação entre a distribuição dos canais da linha lateral e a sua funcionalidade é pouco conhecida para os elasmobrânquios, especialmente para as raias neotropicais da família Potamotrygonidae. A distribuição espacial dos canais e seus túbulos e a quantificação dos neuromastos foram analisadas em exemplares preservados das raias Potamotrygon motoro, P. orbignyi, Potamotrygon sp. "cururu" e Paratrygon aiereba, provenientes do médio rio Negro, Amazonas, Brasil. Foram identificados os canais hiomandibular, infra-orbital, linha lateral posterior, mandibular, nasal e supra-orbital, e os poros e neuromastos de cada um foram quantificados. Nossos resultados mostram que os canais dorsais podem ter a função de identificar a presença de predadores ou presas posicionadas acima do corpo da raia, enquanto os canais ventrais provavelmente permitem uma localização precisa da posição de presas sob o corpo. A existência de canais sem poros na região ventral pode estar relacionada com a alta concentração de neuromastos, que compensaria a falta de contato direto dos neuromastos com o meio externo na detecção de estímulos mecânicos. A concentração dos canais sem poros na proximidade da boca indica sua importância para a localização e captura de presas enterradas no substrato, possivelmente auxiliadas pela presença das vesículas de Savi.

When bacterial cells are stressed by a change in the environment, they respond by changing the activity of enzymes at both the transcriptional and post-transcriptional levels. The UVA component (400-315 nm) of solar radiation reaching the Earth's surface is one of the most common stresses encountered by bacteria in their environment. Bacteria have evolved various antioxidant defense systems to increase survival when subjected to the deleterious effects of UVA irradiation. Recently, UVA-induced cytotoxicity and oxidative damage have been shown to be dependent on radiation intensity and dose distribution, not just total energy dose. We now report that when Escherichia coli is subjected to continuous sublethal, low-fluence UVA irradiation (7.4 W/m(2)) while growing to stationary phase, it responds by changing the activity levels of hydroperoxidases (HPI, HPII), glutathione reductase and manganese superoxide dismutase. This leads to an attenuation of the growth-delay response and an increase resistance to lethal UVA irradiation. When E. coli is given a UVA dose of 135 kJ/m(2) delivered at a fluence rate of 50 W/m(2), extensive protein oxidation occurs which may contribute to the inhibition of key cellular enzymes, leading to cellular dysfunction, DNA damage and eventually death. Changes in antioxidant enzymes induced by low-fluence UVA irradiation do not confer greater protection from protein oxidation after a challenge dose of UVA irradiation delivered at a fluence rate of 50 W/m(2).
